I have built a flow for a process owned by another staff. Now when we send emails from the flow it comes from my email. I want to transfer ownership of the flow so the emails come from the persons email that owns the flow. Can anyone advise how to do this.

Try the first option. Create a new connection. Assign this to the send email step to use. If you don't want to use the other connection anymore you can delete the other connection by clicking the manage connections.

Adding another owner allows others to edit, update and delete the flow. Once the new owner is assinged user need to change the email connections by editing the flow or click the Edit link from the Connection tab.

Do i need to remove the original owner connection for Office 365 outlook and add the new owner connection, or do i just leave the original owner connection active and add the new owner connection?

You could edit the existing connections from the tab or edit the flow step email connections.
