Yes that should work. Are you having a particular issue with the flow you presented? YOu might also want to take a look at htis video. Trigger conditions are now available that can limit when a Flow is actually triggered.
What type of field are you testing? Try changing the NULL to an "" to test for an empty string rather than a null.
Try this as trigger condition
in my case column RunFlow is a single line of text field
This expression will trigger Flow only when RunFlow column has a value

Yes, but what data type is the column in the SharePoint list. Some of the column types will never be null. For example, a string will normally be an empty string and an integer will be zero. If you test those for null it will fail
