Hello,
We have a Sharepoint Online document library with a large amount of items. For future items, there is a metadata field that we would like to make as a required entry. Will changing the field to Required in the document library settings interrupt any documents currently in the Flow associated with this library?

If the flow is created for the list before the field is set to required, it wouldn't affect the flow.

It depends on what the Flow is doing, but I just did a quick test and had no issues. I created a flow that updates a column in a library. The library has two additional text columns. I ran the Flow which updated the first column and it ran with success. I then made the second column required and ran the Flow again. Even though I did not supply data fro the required column, my Flow ran without error. That said, I would run some tests on another library before you implement the required column.
